Following her performance at the Somerset music event on Sunday (June 29), fans took to Twitter to speculate over whether the 9 to 5 hitmaker had lip-synched.
Parton has now broken her silence on the matter, setting the record straight during an interview with The Sun newspaper.
When asked about the miming accusations, the 68-year old hit back: "My boobs are fake, my hair's fake, but what is real is my voice and my heart."
A spokesperson for Dolly was also quick to slam the rumours, stating after her stint on stage: "She sings live. Some people don't know an amazing singer when they hear one."
Lip-synching rumours aside, Glasto co-organiser Emily Eavis has revealed that Parton attracted the biggest crowd she's even seen at the festival.
Eavis told Glastonbury Festivals: "Oh Dolly was the biggest crowd I've ever seen here. She is just a truly amazing live performer.
"Her voice was incredible. And she wrote that song about mud in the morning before the show! I don't think you can beat that performance as a perfect moment here at Glastonbury."
